| Purpose: | PBMCs were isolated by Ficoll from LRS chambers obtained from the Stanford Blood center, then cryopreserved in FBS+10% DMSO in the vapor phase of liquid nitrogen until use. PBMC aliquots were thawed, washed, stained with the HIMC surface phenotyping 33 marker CyTOF panel, EQ normalization beads were added, and then were run on a Helios mass cytometer using the default settings. The resulting Raw data files were normalized separately in two ways: MATLAB (Finck et al) and Fluidigm-ver2. | ||||
