Purpose: To establish whether DRB treatment impacts on the cell cycle and DNA replication in WT U2-OS cells, and the kinetics of these effects.
Conclusion: G1 accumulation and reduction in DNA replication following 100 uM DRB treatment for 6 hours.
Comments: Cells were treated for 0,1,2,4 or 6h with 100 uM DRB (transcription inhibitor). 30 min prior to fixation, cells were incubated with 10 uM EdU, a nucleotide analogue which is actively incorporated into actively replicating cells.
