Purpose: Assessment of reactive oxygen species (ROS) using H2DCFDA stained cells in the presence or absence of cisplatin-mediated stress. PEX5 and PEX19 were overexpressed and compared to control vectors.
Conclusion: PEX5 and PEX19 each reduced ROS steady state levels after cisplatin treatment

Quality control: Unstained cells were used to determine basal green-channel fluorescence for all constructs and cell lines. Propidium Iodide was used to gate out dead cells, as was size selection of small apoptotic blebs. Standard methods were used to gate out doublets.
