Purpose: We aimed to investigate heterogeneity of the human CD8+ memory T cell pool at the protein level, and to confirm at the protein level the findings we obtained by scRNA-Seq. Thus we designed a high-dimensional flow-cytometry panel based on the cluster signature markers CCR7, LEF1, CD161, GZMB and GZMK. This panel was also equipped to detect memory and effector differentiation markers (CD27, CD28, CD45RO, CD127 and T-bet), activation markers (CD38 and HLA-DR), inhibitory receptors (PD-1 and TIGIT) and markers of tissue residency (CD69 and CD103) in CD8+ T cells isolated from the peripheral blood (PB) and tissues like bone marrow (BM), lymph nodes (LN) and lung.
