| Purpose: | To characterize the cytokine profile of T cells in post-COVID-19 subjects using non-specific stimulation (PMA/ionomycin), providing insights into their functional capacity and immune dysregulation. | ||||
| Conclusion: | Analysis of broad functionality in CD8+ T cells revealed increased cytokine production in restrictive lung disease post-COVID19 compared to vaccinated controls. Cytokine profiles were dominated by co-expression of IFN-g and TNF-a. | ||||

| Quality control: | All spectral flow cytometry data were pre-processed by spectral unmixing with autofluorescence subtraction and spill-over correction (SpectroFlow, Cytek Biosciences, Fremont, CA, USA). Removal of dead cells, debris doublets and atypical events (antibody aggregates) was performed by expert gating. Similarly, expert gating was used to pre-gate for CD3+ lymphocytes | ||||
