Notice: Temporary Suspension of New Experiment Creation
We have temporarily disabled the creation of new experiments as we are continuously running out of space. This issue has been impacting both uploads and downloads from FlowRepository. By taking this step, we aim to make downloads of existing data more reliable.
We apologize for the inconvenience and appreciate your understanding as we work on upgrading our hardware and improving the overall solution.

Experiment Overview

Repository ID: FR-FCM-ZY7E Experiment name: Fitness of 14 TDH3 promoter variants in YPD MIFlowCyt score: 49.25%
Primary researcher: Fabien Duveau PI/manager: Patricia Wittkopp Uploaded by: Fabien Duveau
Experiment dates: 2014-04-19 - 2016-04-10 Dataset uploaded: Jun 2017 Last updated: Aug 2017
Keywords: [Gene expression] [fitness optimum] [robustness. ] Manuscripts:
Organizations: University of Michigan, Ann Arbor, MI (USA)
Purpose: Determining the fitness consequences of changes in the expression level of the yeast gene TDH3.
Conclusion: The relationship between TDH3 expression level and fitness in YPD is non-linear and presents a plateau of maximal fitness located around the wild type level of expression.
Comments: Fitness was quantified by competition between each tested strain expressing YFP against a common competitor expressing GFP. The relative frequency of YFP and GFP cells present in each population was recorded at four time points.
Funding: European Molecular Biology Organization postdoctoral fellowship (EMBO ALTF 1114-2012), National Science Foundation (MCB-1021398) and National Institutes of Health (R01GM108826 and 1R35GM118073) grants.
Quality control: The FCS data were collected from seven plates of 96 samples scored at four different time points and in 3 separate experiments including one or three plates each. Each plate contains 24 replicates of the control YFP strain with a wild type copy of TDH3 competed against the GFP competitor. These controls can be used to correct for variation existing between plates.
