Purpose: The purpose of the experiment was to determine sensitivity of RNA flow cytometry by analyzing samples with low abundant intracellular RNA. Slides made from the suspension stained samples were run by flow and were also imaged on slides. MFI from the flow analysis was correlated to RNA spot count from the imaging analysis to determine sensitivity.
Conclusion: It was determined from this experiment that it is possible to distinguish cells containing only 1-2 spots by flow cytometry, by correlating MFI values of gated cells to the spot counts generated by image analysis.
