Purpose: This panel is an expansion and optimization of the OMIP-014 panel. It was developed, optimized and qualified for the assessment of human antigen-specific T-cell responses by intracellular cytokine staining, including memory markers and Tfh cells. Additionally NK-cell responses are also evaluated. The functional markers assessed include IFNg, IL-2, TNFa, IL-4 and IL-21.
Conclusion: The panel allows successfully the assessment of antigen-specific CD4+ and CD8+ T cell responses, as well the evaluation of circulating Tfh, the characterization of memory differentiation of the cytokine producing cells and the detection of NK cells responding to antigen stimulation. The panel underwent assay qualification and proved to be as sensitive for IFNg, IL-2 and TNFa as the validated OMIP-014 panel.
