Purpose: | To demonstrate application of DNA ploidy analysis using Fxcycle violet (FCV) and simultaneous multicolor (4-7) surface immunophenotyping. | ||||
Conclusion: | This method DNA ploidy analysis using FxCycle violet and simultaneous six color immunophenotyping can be used flow cytometric DNA ploidy determination in B-ALL MRD evaluation (up to 100 events). | ||||
Comments: | This experiment proves that this method can be useful for DNA ploidy analysis for B-ALL MRD analysis. However, it is recommended acquire at least 200 events of tumor cells for good results for rare events of tumor. Although, CV of G1 peak might be slightly higher (between 4 to 6), it is acceptable as it is still falls within the 10% standard error and does not affect the results of the assay. Few unstained and apoptotic cells always present in the sample showing decreasing staining pattern of FxCycle. However, these does not interfere with analysis and interpretation once gated out. | ||||

Quality control: | DNA ploidy and simultaneous six color immunophenotyping using FxCycle-Violet. | Daily routine QC as per manufacturer's instructions and compensation (both generic and tube specific post-acquisition) was performed.
